All great tips, but for the most part I think the mix tape should reflect the real you not something with a huge production behind it. I feel using a DAW as a sequencer is cheating because it is not you doing the mix; your computer is doing it. However, it is valid to use it to master its sound and to edit the length of each song so that it does not become boring.
I guess in the end it depends on who you are making the mix tape for. You may want to show you can play (mix) different music genres if the club you are “auditioning” for plays different genres, or you may want to mix the tunes that really appeal to you if you want to establish your name and style, and you may want to include fun silly songs if you are trying for a wedding type event. As with most mucicians in an audition, the beggining of your presentation will tell your listener if he wants to continue with it or stop it and go to the next one; make sure it is impresive enough to catch their attention.
Finally I would suggest to make sure you enjoy making and listening to that mix tape and hope that other people enjoys it as much as you do.
